2DLC-MS/MS method

The 2DLC-MS/MS method is effective for global analyses of complex mixed protein samples.

Peptide separation by the 2DLC method - Peptide separation by ZIC-HILIC -
In global analyses, mixed protein samples are analyzed. Therefore, peptides should be separated by two-dimensional LC before they are analyzed by mass spectrometry.

ZIC®-HILIC method

The stationary phase of ZIC®-pHILIC (porous methacrylate beads bound to zwitterionic groups) has strong retention of hydrophilic compounds. In addition, it is characterized by weak electrostastic interactions between the zwitterion groups of the stationary phase and the positive or negative charges of the sample. Separation by HILIC mode can increase the composition ratio of the organic solvent in the mobile phase and decrease its salt concentration; thus, inhibitory effects on peptide absorption can be expected.

ZIC®-pHILIC also has excellent operability, for example, in concentration of fractionated samples.

A wide range of conditions can be set, and selectivity can be changed in diverse ways. In addition, since the column is polymer-based, it allows setting of a wide pH range (pH2-12).
